[Rationale for the Classification] Although there was no data on this substance itself, based on (1) and (2), this insoluble substance was considered to have the same effects as soluble nickel compounds on a living body after being taking into the body, and it was classified in Category 2 for this hazard class as is the case with soluble nickel compounds. [Evidence Data] (1) Nickel compounds are not mutagenic in bacteria and are only weakly mutagenic in cultured mammalian cells under standard test conditions but can induce DNA damage, chromosomal aberrations, and micronuclei formation in vitro and in vivo (IARC 100C (2012)). (2) Both soluble and insoluble nickel compounds have the potential for giving rise to DNA breaks, DNA-protein crosslinks, and chromosomal damage in vivo and in vitro (ECHA RAC Opinion on scientific evaluations of OELs (2018)). [Reference Data, etc.] (3) Crystalline nickel sulfides are phagocytized by a large variety of different cells in culture (IARC 100C (2012)). (4) In an inhalation exposure test for trinickel disulfide (CAS RN 12035-72-2) with rats for 1 or 4 weeks (0.04 to 0.60 mg/m3, 6 hours/day, 5 days/week), activation of genes related to genotoxicity in lung tissue was observed in two highest dose groups at or above 0.15 mg/m3 (ECHA RAC Opinion on OEL Evaluation (2018)).
