[Rationale for the Classification] It was classified in Category 2 from (1)-(4). Besides, knowledge in humans was prioritized although it is also reported that it was not irritating in tests in rabbits or rats (7), (8). The category was revised by using new information sources. [Evidence Data] (1) It is reported that this substance is irritating to the skin, eye, and mucous membranes (ACGIH (2005), NICNAS IMAP (Accessed Dec. 2018), DFGOT (2004)). (2) It is reported that erythema, pain, and dryness of the skin occur when this substance contacts the eye or skin (Environmental Risk Assessment for Chemical Substances Vol. 7 (Ministry of the Environment, 2009)). (3) It is reported that irritation was observed after occlusive or open application of this substance on six humans (NICNAS IMAP (Accessed Dec. 2018)) (4) There are multiple reports that it was irritating in skin irritation tests in rabbits, guinea pigs, rats, or mice (SIAR (2000), Environmental Risk Assessment for Chemical Substances Vol. 7 (Ministry of the Environment, 2009), NICNAS IMAP (Accessed Dec. 2018)). [Reference Data, etc.] (5) In the announcement No. 33 of the Ministry of Labour (1996) (revised by the announcement No. 316 of the Ministry of Health, Labour and Welfare, 2013), this substance was designated as "tetrahydrofuran" in Simple chemical substances or compounds designated by the Minister of Health, Labour and Welfare based on Appended Table 1-2, (iv) 1 of the Ordinance for Enforcement of the Labor Standards Act, and a disease principally having specific symptoms and disorders (subjective symptoms such as headaches, dizziness, and vomiting or skin disorders) due to work involving exposure to this substance was designated as an occupational disease by the Minister of Health, Labour and Welfare. (6) In the notice No. 0811001 issued by Director of Labour Standards Bureau, the Ministry of Labour (2003), this substance was designated as "tetrahydrofuran" among hazardous substances for which installation of protective equipment to prevent skin disorders is mandated in Article 594 in the Industrial Safety and Health Regulations. (7) It is reported that in a skin irritation test with rabbits (n = 6), after 72-hour occlusive application of this substance, PII: 1.93, erythema score: 0.75, and edema score: 0.9 were obtained after 72 hours, and it was not irritating (ACGIH (2005), REACH registration dossier (Accessed Dec. 2018)). (8) It is reported that in an acute dermal toxicity test with rats (OECD TG402, n = 5 (males), 5 (females)), after 24-hour semi-occlusive application of this substance (purity > 99.8%), no erythema or edema was found (REACH registration dossier (Accessed Dec. 2018)). |
