Based on reports of LC50 values for mice of 2,300 ppm (converted 4-hour equivalent value: 3,043 ppm) (DFGOT Vol.25 (2009)) and 3,000 ppm (converted 4-hour equivalent value: 3,969 ppm) (ACGIH (7th, 2001)) in 7-hour inhalation exposure tests, and an LC50 value for mice of 2,043 ppm (converted 4-hour equivalent value: 2,889 ppm) in an 8-hour inhalation exposure test (DFGOT vol. 25 (2009)), it was classified in Category 4. There is a description that contact with human skin causes irritation and burns, and long-term contact causes drying and cracking (HSDB (Access on August 2017)). As for rabbits, it is described that burns and denaturation were observed in a skin irritation test (HSDB (Access on August 2017)), and that tissue damage occurred in another occlusive application test, and degreasing of the skin was present in a non-occlusive application test (DFGOT vol. 25 (2009)). In a combined repeated dose toxicity study with the reproduction/developmental toxicity screening test (OECD TG 422) with rats dosed by gavage, out of 12 rats, 2 did not mate and 2 became sterilized at the maximum dose (300 mg/kg/day) at which general toxic effects on the liver, kidney, etc. were observed in male and female parental animals. Moreover, extension of the pregnancy period, high rate of stillbirth, low value of live birth index and a low value trend of the 4-day survival rate of the newborn pups were observed (JECDB (Access on August 2017)). In addition, it was described that as the result of inhalation exposure at 1,010 ppm for 114 days in male rabbits or guinea pigs, reduced spermatogenesis and tubular fibrosis in the testes occurred, but the number of animals used is insufficient to use for an evaluation (DFGOT Vol. 25 (2009)). From the above, since developmental effects were observed at the level where general toxicity occurred in maternal animals in the combined test with rats, it was classified in Category 2. Besides, classification became possible by using new information after the previous classification.

As for humans, it was reported that during use of this substance as a fire extinguishing agent, three firefighters, who were exposed to the vapor or sprayed liquid of this substance for a short time, showed severe headache, nausea, burning sensation in the stomach, and vomiting, and hyperemia in the mucosa of the upper respiratory tract and conjunctiva were observed. There is a description as follows: in two out of the three firefighters, who were exposed close to the fire extinguishing point, disorientation, unconsciousness and generalized spasms etc. were observed and the other firefighter, who was exposed at some distance from the point, did not develop disorientation or unconsciousness but complained of headache for 3 months after the exposure; however, all three recovered (Environmental Risk Assessment for Chemical Substances Vol.6, Tentative Hazard Assessment Sheet (Ministry of the Environment, 2008), ACGIH (7th, 2001), DFGOT vol. 25 (2009), PATTY (6th, 2012)). As for experimental animals, it was reported that in a 15-minute single inhalation exposure test with rats, hypersensitivity, an impaired sense of balance and convulsive tremors were observed at or above 1,500 ppm (DFGOT vol. 25 (2009)). In addition, it was described in ACGIH (7th, 2001) that in the same 15-minute inhalation exposure test with rats, a slight narcotic effect was observed at 3,000 ppm. From the above information on humans and experimental animals, it was classified in Category 1 (central nervous system) and in Category 3 (narcotic effects). Since generalized spasms were observed in humans, it was classified in Category 1 (central nervous system), and the classification result was different from the previous one.

As for experimental animals, in a combined repeated dose toxicity study with the reproduction/developmental toxicity screening test (OECD TG 422) with rats dosed by gavage, erosion of the glandular stomach, hypertrophy of the centrilobular hepatocytes, single cell necrosis and micro granuloma, etc. of hepatocytes were observed at or above 100 mg/kg/day (converted guidance value: 46.7 mg/kg/day), which is within the guidance value range for Category 2, and increases of relative weight of the liver, spleen, and adrenal gland, vacuolation of the adrenal fasciculata, vacuolation or necrosis of the proximal tubules, basophilic change of renal tubules and granulation of epithelial cell of the papillary duct in the kidney were reported at 300 mg/kg/day (converted guidance value: 140 mg/kg/day) which is beyond the guidance value range for Category 2 (JECDB (Access on August 2017)). In addition, several inhalation tests were performed, and effects were reported at doses exceeding the guidance value range of Category 2, but these doses were mostly the minimum dose for the test, and the effects at doses lower than the minimum dose were unknown (Environmental Risk Assessment for Chemical Substances Vol.6, Tentative Hazard Assessment Sheet (Ministry of the Environment, 2008)).
From the above, it was classified in Category 2 (liver).
Besides, the classification result was different from the previous one due to the use of a new information source.
