In a combined repeated dose toxicity study with the reproduction/developmental toxicity screening test (OECD TG 422) with rats dosed by gavage, slight reproductive and developmental effects such as low trends of birth rate, delivery rate, and implantation rate, a decrease in the number of offspring born, and decreases in the number of births and the number of live pups on postnatal day 4 in pups were observed at the high dose (500 mg/kg/day), but they were effects at the dose where severe toxicities (death (3/12 cases), histopathological changes in the forestomach and femurs) were observed in maternal animals (Safety Test (Ministry of Economy, Trade and Industry (METI, 2007))). On the other hand, in a continuous breeding test with mice dosed by feeding, in parental animals, a low value of body weight of maternal animals and decreased litter size at doses of the mid dose (0.75% (1,125 mg/kg/day)) or above, a low value of body weight of paternal animals, a decreased fertility rate and extension of days until delivery in the third gestation to the fifth gestation at the high dose (1.5% (2,250 mg/kg/day)), and in pups, at the high dose, a low value of pre-weaning viability index in F1 and a decreased number of live pups in F2 were observed (NTP Report for RACB88105 (1990)).
From the above, since effects on fertility (decreased fertility, decreased litter size, etc.) and a low value of viability index in pups were observed in the NTP continuous breeding test with mice at doses where general toxicity (a low value of body weight) was manifested in parental animals, it was classified in Category 2 for this hazard class.

As for experimental animals, in a combined repeated dose toxicity study with the reproduction/developmental toxicity screening test (OECD TG 422) with rats dosed by gavage, squamous hyperplasia of the forestomach at 100 mg/kg/day (converted guidance value: 47 mg/kg/day) within the guidance value range for Category 2, and salivation, decreased body weight gain, low value tendency of the erythrocyte count, hemoglobin concentration and hematocrit value, decreased testes weight, erosion and focal inflammatory cell infiltration of the forestomach, an increase in the trabecular bones in the femur, degeneration in the seminiferous epithelium in the testes, decreased sperm in the epididymal duct, etc. at 500 mg/kg/day (converted guidance value: 233 mg/kg/day) exceeding the guidance value range for Category 2 were observed (Safety Test (Ministry of Economy, Trade and Industry (METI, 2007))).
From the above, lesions of the forestomach observed within the guidance value range for Category 2 are thought to be due to irritation. Therefore, it was classified as "Classification not possible."
