Rationale for the classification
1 Acute toxicity (Oral) Category 4

Warning
H302 P301+P312

P270
P330
P501
Four data of LD50 values for rats of 900 mg/kg, 900 mg/kg, 3,000 mg/kg, and 3,200 mg/kg are reported (all from PATTY (6th, 2012)). Two of these correspond to Category 4 while the other two correspond to "Not classified" (Category 5 in the UN GHS classification). By adopting the category with higher hazard, it was classified in Category 4.
1 Acute toxicity (Dermal) Not classified
-
-
- - Based on a reported LD50 value of > 20 mL/kg (> 20,600 mg/kg from a specific gravity of 1.03) for guinea pigs (PATTY (6th, 2012)), it was classified as "Not classified."
1 Acute toxicity (Inhalation: Gases) Not applicable
-
-
- - Liquid (GHS definition)
1 Acute toxicity (Inhalation: Vapours) Classification not possible
-
-
- - Classification not possible due to lack of data. There is a report that there were no death cases in a test in which rats were exposed by inhalation to the saturated vapour (approx. 430 ppm, 2.11 mg/L) of this substance for 8 hours (PATTY (6th, 2012), ACGIH (7th, 2009)). However, the category could not be specified on the basis of this data alone. Therefore, it was classified as "Classification not possible." Besides, since the exposure concentration was lower than 90% of the saturated vapor pressure concentration (524 ppm), a reference value in the unit of ppm was applied as vapour with little mist.
1 Acute toxicity (Inhalation: Dusts and mists) Classification not possible
-
-
- - Classification not possible due to lack of data.
2 Skin corrosion/irritation Category 2

P321
Based on a description that this substance is irritating to the skin (PATTY (6th, 2012), HSDB (Access on June 2017)), it was classified in Category 2. The category was revised based on information obtained in this survey.
3 Serious eye damage/eye irritation Category 2A

Warning
H319 P305+P351+P338
P337+P313
P264
P280
Based on a description that severe irritation was observed in an evaluation 24 hours after an application of this substance in an eye irritation test with rabbits (PATTY (6th, 2012)), it was classified in Category 2A. Besides, this substance was classified as "Eye Irrit. 2" in the EU CLP classification (ECHA CL Inventory (Access on June 2017)).
4 Respiratory sensitization Classification not possible
-
-
- - Classification not possible due to lack of data.
4 Skin sensitization Classification not possible
-
-
- - Although it was reported that both of two skin sensitization tests with guinea pigs (the modified Draize method and the maximization method) were negative (PATTY (6th, 2012), ACGIH (7th, 2009)), it was classified as "Classification not possible" because no data were obtained in humans. The category was changed from the previous classification since the contents of information sources were reviewed.
5 Germ cell mutagenicity Classification not possible
-
-
- - Classification not possible due to lack of data. There were no in vivo data. As for in vitro, a bacterial reverse mutation test was negative (IRIS (1988), PATTY (6th, 2012)).
6 Carcinogenicity Classification not possible
-
-
- - No test data was available in either humans or experimental animals. However, since the EPA classified it in Group D (IRIS (1988)), it was classified as "Classification not possible."
7 Reproductive toxicity Category 2

When this substance was administered by gavage to female rats from 14 days prior to mating to day 3 of lactation, in maternal animals, as toxic symptoms, urine stains, a decrease in locomotor activity, pale skin, rough fur, etc. at or above 225 mg/kg/day, and unsteady gait at 750 mg/kg/day were observed. As for effects on the pups, a decrease in the live birth index and decreased number of alive pups by day 4 of lactation were observed (ACGIH (7th, 2009), Environmental Risk Assessment for Chemical Substances Vol.7, Tentative Hazard Assessment Sheet (Ministry of the Environment, 2009)). From the above, it was classified in Category 2 for this hazard class since effects on the next generation were observed at the dose where maternal toxicity occurred.

With regard to humans, it is described that this substance was used as an anesthetic in the 19th century (ACGIH (7th, 2001)). As for experimental animals, central nervous system depression was reported to be a toxic symptom in a single oral dose test with rats (ACGIH (7th, 2009), PATTY (6th, 2012)). As for the inhalation route, it was reported that in a test with mice to evaluate the irritation potential of this substance, when a decline in the respiration rate following short-duration (5 minutes) inhalational exposure to the vapor was measured, an RD50 value was 500 mg/m3 (ACGIH (7th, 2009), PATTY (6th, 2012)). From the above, it was classified in Category 3 (respiratory tract irritation, narcotic effects). Besides, it was reported that in a test with mice, administration by subcutaneous or intravenous injection showed hypnotic effects, and in intraperitoneal administration, clear hypnotic effects were observed at 400-500 mg/kg (ACGIH (7th, 2009)).
9 Specific target organ toxicity - Repeated exposure Classification not possible
-
-
- - No information on humans is available.
As for experimental animals, in a 17-week repeated toxicity test with rats dosed by feeding, no effects were observed even at 1% (0.845% due to volatilization (423 mg/kg/day)) exceeding the guidance value range for Category 2 (Environmental Risk Assessment for Chemical Substances Vol.7, Tentative Hazard Assessment Sheet (Ministry of the Environment, 2009), ACGIH (7th, 2009), IRIS (1988), PATTY (6th, 2012)). Also, it is reported that a 4-week repeated toxicity test with rats dosed by gavage, hyaline droplet nephrosis in the males at or above 75 mg/kg/day (converted guidance value: 23 mg/kg/day) within the guidance value range for Category 2, urine stains and salivation at or above 225 mg/kg/day (converted guidance value: 70 mg/kg/day), and a low value of body weights, reduced forelimb grip strength, a decrease in locomotor activity, and unsteady gait at 750 mg/kg/day (converted guidance value: 233 mg/kg/day) exceeding the guidance value range for Category 2 were reported (Environmental Risk Assessment for Chemical Substances Vol.7, Tentative Hazard Assessment Sheet (Ministry of the Environment, 2009), ACGIH (7th, 2009)). Other than these, it was reported that in a 14-day inhalation toxicity study with mice exposed to the vapor, no effects on the trachea were observed at 300 ppm (converted guidance value: 0.23 mg/L) within the guidance value range for Category 2 (PATTY (6th, 2012)).
From the above, effects on the nervous system were reported, but these were observed at a dose exceeding the guidance value range for Category 2.
Therefore, it was classified as "Classification not possible."
Besides, since new information sources were used, the classification result was changed from the previous classification.
10 Aspiration hazard Classification not possible
-
-
- - Classification not possible due to lack of data.

Rationale for the classification
11 Hazardous to the aquatic environment (Acute) Not classified
-
-
- - From 96-hour LC50 = 162 mg/L for fish (Pimephales promelas) (ECETOC TR91: 2003), it was classified as "Not classified."
11 Hazardous to the aquatic environment (Long-term) Not classified
-
-
- - Reliable chronic toxicity data were not obtained. Due to being rapidly degradable (readily biodegradable, a degradation rate by BOD: 64.7% (J-CHECK, 1977)), no bioaccumulation (LogKow: 1.58 (PHYSPROP Database: 2017)), and "Not classified" in acute toxicity, it was classified as "Not classified."
12 Hazardous to the ozone layer Classification not possible
-
-
- - No data available.
