Besides, it is described that chlorinated naphthalenes are hepatotoxic (CICAD 34 (2001), DFGOT vol. 13 (1997)) and that symptoms described in workers exposed to chlorinated naphthalenes included irritation of the eyes, fatigue, headaches, anaemia, haematuria, anorexia, vomiting, and severe abdominal pain (CICAD 34 (2001)). There is no information however on the frequency of exposure in each case. In the previous classification, the substance was classified in Category 2 (systemic toxicity) based on the description that "Symptoms of exposure to this substance may include, headaches, fatigue, vertigo and anorexia" (HSDB (2003)). However, the frequency of exposure was not clear, and it was not possible to refer to original information sources. Therefore, the classification was changed. |

As for experimental animals, in a 28-day repeated dose toxicity test using rats administered by gavage, a decrease in motor activity, crouching position, lower body weights (males), an increase in relative liver weights (females) and centrilobular hypertrophy of hepatocytes were observed at 100 mg/kg/day or above (converted guidance value: 31 mg/kg/day), which is equivalent to Category 2 (JECDB (Access on October 2016)). Moreover, higher value in ALT (males), gamma-GT (males) and absolute and relative liver weights etc. were observed at 300 mg/kg/day (converted guidance value: 93 mg/kg/day) (JECDB (Access on October 2016)). In the previous classification, the substance was classified in Category 2 (skin) based on the description in HSDB that "chloracne may develop after long term exposure" (HSDB (2003)). However, there was no description that chloracne was observed as a case of this substance, and it was described that chloracne was observed after exposure to trichloronaphthalenes and higher chlorinated naphthalenes (DFGOT vol. 13 (1997)). Therefore, skin was not adopted as the target organ. From the above, the substance was classified in Category 2 (liver). |
