There are no epidemiological data available for classification on human carcinogenicity related to exposure to this substance. As for experimental animals, there is a description that as a result of subcutaneous administration of the hydrochloride of this substance (25-100 mg/kg, once/week, 30 weeks) to rats, occurrences of tumors in the brain, olfactory bulb, mammary gland, and liver were observed, and there is a description that in an experiment in which pregnant rats were given a single intravenous dose of this substance on day 15 of gestation (50, 150 mg/kg), most of the F1 pups died from tumors of the brain, spinal cord or peripheral nervous system on 126-498 days after birth (IARC 4 (1974)).
Based on these test results, IARC classified it in "Group 2B" (IARC Suppl. 7 (1987), IARC 71 (1999)). As for classification results by other organizations, Japan Society for Occupational Health (JSOH) classified it in "Group 2B" (Recommendation of Occupational Exposure Limits (2015)). From the above, it was classified in Category 2 for this hazard class.

As described in hazard class of carcinogenicity, there is a description that as a result of single intravenous administration at doses of 50 and 150 mg/kg to pregnant rats on day 15 of gestation, tumors of the cranial nervous system (brain, spinal cord, peripheral nervous system) were observed in pups (IARC 4 (1974)). In addition, there are the following descriptions in Catalog of Teratogenic Agents: When pregnant rats were given a single dose of 12-33% of an LD50 value on day 15 of gestation, nervous system tumors in F1 pups occurred at high frequency in any route, but no tumors occurred in the administration on the day before day 11 of gestation. On the other hand, it is described that in administration of 10-15 mg/kg on day 10 of gestation, developments of the eyes and optic nerve were completely inhibited, and in administration in late gestation, hydrocephalus and malformations of the limbs were caused (Catalog of Teratogenic Agents. (13th 2010), HSDB (Access on October 2015)).
From the above, when this substance is administered to pregnant rats, it causes nervous system tumors in the next generation, and the findings were obtained that suggest the possibility that disturbance of organogenesis and malformations in fetuses are induced depending on the condition setting such as the timing of administration during the gestational period and the doses, and although the route of administration was the route by injection, induction of malformations and development of tumors occurred only by a single exposure in the embryonic phase, therefore, it is considered that these effects can sufficiently occur by usual exposure routes. Thus, it was classified in Category 1B for this hazard class.
