- | - | In a carcinogenicity test in which this substance dissolved in ethanol was dermally applied for 2 years (5 days/week) in rats or mice, increased incidences of tumors were not observed after administration up to the doses where inflammations and ulcers in forestomach and degeneration in liver in male rats and deaths and so on in female rats occurred and to the doses where deaths, ulcer in forestomach, and epidermal hyperplasia at application sites in male mice and degeneration, necrosis, and fibrosis of cardiac muscles and so on in male and female mice occurred. There is the comment that there is no evidence of carcinogenicity in either males or females of rats and mice under the conditions of this test (NTP TR536 (2011)). Therefore, it is conceivable that it corresponds to "Not classified" in a dermal route, but information on carcinogenicity in other routes and an epidemiological survey in humans is lacking. As to classification by other organizations, there is only the classification result of D (not classifiable as to human carcinogenicity) by US EPA (IRIS Summary (Access on July 2015)). There is no human information. As for experimental animals, in a 14-week (70 days) dermal administration toxicity test using rats, at 200 mg/kg/day (converted to a 90-day equivalent: 156 mg/kg/day) that is within a range of Category 2, vacuolization of muscle fiber cells and interstitial mononuclear cell infiltration in the heart, and necrosis of muscle fiber in the heart were observed (NTP TR536 (2011)). Moreover, in a 105-week dermal administration toxicity test using rats, at 150 mg/kg/day that is within a range of Category 2, degeneration of olfactory epithelium and inflammation of forestomach were found (NTP TR536 (2011)). As above, effects on the heart, respiratory organs, and digestive tract were observed, but forestomach was not included in target organs because human does not have one. Therefore, the substance was classified in Category 2 (heart, respiratory organs). |
