Rationale for the classification
1 Acute toxicity (Oral) Not classified
-
-
- - It was classified as "Not classified" (Category 5 in UN GHS classification) based on LD50 values for rats of 2,490 mg/kg (ACGIH (7th, 2001), PATTY (6th, 2012)) and 3,690 mg/kg (ACGIH (7th, 2001)). Besides, data in PATTY (6th, 2012) obtained in this investigation were added, and the classification was changed from Category 5 to "Not classified" according to the Classification JIS.
1 Acute toxicity (Dermal) Not classified
-
-
- - It was classified as "Not classified" based on an LD50 value of > 5,000 mg/kg for rabbits (PATTY (6th, 2012), IUCLID (2000)).
1 Acute toxicity (Inhalation: Gases) Not applicable
-
-
- - Liquid (GHS definition)
1 Acute toxicity (Inhalation: Vapours) Classification not possible
-
-
- - Classification not possible due to lack of data.
1 Acute toxicity (Inhalation: Dusts and mists) Classification not possible
-
-
- - Classification not possible due to lack of data.
2 Skin corrosion/irritation Category 2

Warning
H315 P302+P352
P332+P313
P362+P364
P264
P280
P321
It was classified in Category 2 because it is described that it is irritating to the skin, eye, and respiratory tract (HSDB (Access on September 2013)), and it was moderately irritating in rabbits and humans in a GLP test according to the EC guideline (IUCLID (2000)).
3 Serious eye damage/eye irritation Category 2

Warning
H319 P305+P351+P338
P337+P313
P264
P280
It is described that it causes irritation to the eye (IARC (1986)), and it is irritating to the eye, and blepharospasm was observed in an eye irritation test with rabbits (HSDB (Access on September 2013)). Because there are no descriptions of reversibility, it was classified in Category 2 based on the above information.
4 Respiratory sensitization Classification not possible
-
-
- - Classification not possible due to lack of data.
4 Skin sensitization Classification not possible
-
-
- - It is described in ACGIH (7th, 2001) that no reaction was observed at an 8% concentration of this substance in a sensitization test in volunteers, and it is described that it was negative in a Buehler test and a maximization test with guinea pigs and negative in a patch test in humans (IUCLID (2000)), but there is a description that sufficient data on sensitization were not available (ACGIH (7th, 2001)). Therefore, it was classified as "Classification not possible."
5 Germ cell mutagenicity Classification not possible
-
-
- - It was classified as "Classification not possible" because it was not possible to classify a substance as "Not classified" according to the revised GHS classification guidance for the Japanese government. As for in vivo, it was negative in all of micronucleus tests with bone marrow cells and peripheral blood erythrocytes of mice (IARC (1999), JECFA TRS 909 (2002), NTP DB (Access on June 2013), IUCLID (2000)), a chromosomal aberration test with mouse bone marrow cells (IARC (1999), JECFA TRS 909 (2002), NTP DB (Access on June 2013)), a sister chromatid exchange test with mouse bone marrow cells (IARC (1999), JECFA TRS 909 (2002), NTP DB (Access on June 2013), IUCLID (2000)), an unscheduled DNA synthesis test with rat hepatocytes, and a DNA strand-break test with rat pancreas (IARC (1999), JECFA TRS 909 (2002)). As for in vitro, there are positive data in a mouse lymphoma test with cultured mammalian cells (IARC (1999), JECFA TRS 909 (2002), NTP DB (Access on June 2013)), but it was negative in a bacterial reverse mutation test (IARC (1999), ACGIH (7th, 2001), JECFA TRS 909 (2002), NTP DB (Access on June 2013), IUCLID (2000)) and a chromosomal aberration test with cultured mammalian cells (IARC (1999), JECFA TRS 909 (2002), NTP DB (Access on June 2013)).
6 Carcinogenicity Classification not possible
-
-
- - It was classified as "Classification not possible" because it is classified in A4 in ACGIH (1994) and Group 3 in IARC 71 (1999). The category was changed according to the GHS classification guidance for the Japanese government.
7 Reproductive toxicity Classification not possible
-
-
- - Classification not possible due to lack of data. Besides, as for developmental toxicity, in a developmental toxicity test with rats in the oral (gavage) route, maternal animals showed slightly reduced weight gain, and a decrease in fetal body weights was observed. No malformations were seen, but internal and skeletal variations were found, which were thought to be due to maternal toxicity and lower fetal body weights (JECFA FAS 868 (1996)). Furthermore, it is reported that in 13-week oral administration toxicity tests with rats or mice, no effects on the sperm and genetic organs were seen in males, and extension of the estrous cycle was observed in female mice, but this was associated with lower body weights (JECFA FAS 868 (1996), NTP TR431 (1993)), and it is described in PATTY (6th, 2012) that this substance was not considered to be a reproductive toxin because no compound-related effects on the genetic organs were found in subchronic toxicity tests with rats and mice. However, it was classified as "Classification not possible" because no sufficient tests on fertility effects were conducted.

It was classified in Category 1 (respiratory organs), Category 3 (narcotic effects) because it is described that it produces respiratory tract irritation and narcotic effects in humans (NTP TR250 (1986), ACGIH (7th, 2001)), and lung hyperemia, pulmonary edema (ACGIH (7th, 2001)), and central nervous system depression (NTP TR431 (1993)) were observed in mice after exposure to vapour (1.3 mg/L) within the guidance value range for Category 1.
9 Specific target organ toxicity - Repeated exposure Category 1 (kidney), Category 2 (nasal cavity)

Danger
Warning
H372
H373
P260
P264
P270
P314
P501
It was classified in Category 1 (kidney) because it is described that continued inhalation exposure to this substance results in kidney damage in humans (NTP TR250 (1986)). On the other hand, as for experimental animals, no toxicity effects were seen even after exposure to the doses above the range for Category 2 in a 2-year diet administration test with rats (NTP TR431 (1993)), but it is described that nasal lesions (atrophy and degeneration of the olfactory epithelium, hyperplasia of the nasal submucosal glands, pigmentation of the nasal mucosal epithelium) were observed at the dose (35-40 mg/kg/day) within the guidance value range for Category 2 in a 2-year diet administration test with mice (NTP TR431 (1993)). It was classified in Category 2 (nasal cavity) from the viewpoint of assessing adverse effects while standing on the safe side.
10 Aspiration hazard Classification not possible
-
-
- - Classification not possible due to lack of data.
