Rationale for the classification
1 Explosives Not applicable
-
-
- - There is a chemical group associated with explosive properties (neighboring nitrogen) in the molecule, but it was judged to be not explosive because the group is an aromatic azo group.

9 Pyrophoric liquids Not applicable
-
-
- - Solid (GHS definition)
10 Pyrophoric solids Classification not possible
-
-
- - No data available.
11 Self-heating substances and mixtures Classification not possible
-
-
- - No data available.
12 Substances and mixtures which, in contact with water, emit flammable gases Not applicable
-
-
- - The chemical structure of the substance does not contain metals or metalloids (B, Si, P, Ge, As, Se, Sn, Sb, Te, Bi, Po, At).
13 Oxidizing liquids Not applicable
-
-
- - Solid (GHS definition)
14 Oxidizing solids Not applicable
-
-
- - The substance is an organic compound containing oxygen (but not fluorine or chlorine) which is chemically bonded only to carbon or hydrogen.
15 Organic peroxides Not applicable
-
-
- - Organic compounds containing no bivalent -O-O- structure in the molecule.
16 Corrosive to metals Classification not possible
-
-

Rationale for the classification
1 Acute toxicity (Oral) Classification not possible
-
-
- - Classification not possible due to lack of data.

It is reported in HSDB (Access on October 2013) that allergic dermatitis was induced by wearing stockings for which this substance was used as a dye, and in patch tests with several dyes including this substance in patients with allergic dermatitis, they showed strong reactions to this substance. Furthermore, this substance is classified in "R43" in EU DSD classification and "Skin Sens. 1 H317" in EU CLP classification. Base on the above information, it was classified in Category 1.
5 Germ cell mutagenicity Classification not possible
-
-
- - Classification not possible due to lack of data. As for in vivo, it was negative in a micronucleus test and a chromosomal aberration test with mouse bone marrow cells but positive in a sister chromatid exchange test (NTP DB (Access on August 2013)). As for in vitro, it was negative in a chromosomal aberration test with cultured mammalian cells but positive in a bacterial reverse mutation test and a mouse lymphoma test (NTP DB (Access on August 2013), IARC 48 (1990)). Therefore, it was classified as "Classification not possible" by expert judgment.
6 Carcinogenicity Classification not possible
-
-
- - Because this substance is classified in Group 3 by IARC (IARC (1990)), it was classified as "Classification not possible."
7 Reproductive toxicity Classification not possible
-
-
- - Classification not possible due to lack of data.
8 Specific target organ toxicity - Single exposure Classification not possible
-
-
- - Classification not possible due to lack of data. Besides, it is reported that no toxic signs were observed after oral diet administration to rats even at up to 100,000 ppm (corresponding to 5,000 mg/kg) (NTP TR222 (1982)).
9 Specific target organ toxicity - Repeated exposure Classification not possible
-
-
- - In 13-week or 2-year feeding tests with rats and mice, effects (non-neoplastic changes) on the thyroid (follicle-cell hyperplasia), spleen (hemosiderosis), kidney (pigment deposition in the tubules), and liver (swelling of the centrilobular hepatocytes, foci of vacuolated cells, foci of eosinophilic cells) were observed at the doses (corresponding to 250-750 mg/kg/day) above the guidance value range for Category 2 (NTP TR222 (1982), IARC 48 (1990)). Therefore, it corresponds to "Not classified" in the oral route. However, because of no toxicity information on the other routes, it was classified as "Classification not possible" due to lack of data.
10 Aspiration hazard Classification not possible
-
-
- - Classification not possible due to lack of data.
