In a combined repeated dose toxicity study with the reproduction/developmental toxicity screening test by oral administration to rats (doses: 0, 2.5, 10, 40, 160 mg/kg/day) (OECD TG422, GLP), no effects on estrous cycle, copulation, fertilization, gestation period, and delivery were observed in either group, and for offspring, no treatment-related effects were seen in the number of live pups on day 0 of lactation, the number of stillbirths, sex ratio, delivery index, and live birth index, but all pups of two maternal animals died by day 4 of lactation in the 160 mg/kg dose group, and effects on survival/development of pups were suggested. Furthermore, in the 160 mg/kg dose group, there was a tendency toward an increase in pup deaths during a nursing period and a tendency toward a decrease in viability index on day 4 of lactation (JECDB (Access on May 2012)). Therefore, it was classified in Category 2. As effects in parent animals, a decrease in cytoplasmic lipid droplets in the liver in males at or above 40 mg/kg and a slight increase in glycogen deposits in hepatocytes in females were found. Besides, there is also a report that fetal deaths and an increase in malformations such as cleft lip and polydactyly were observed after subcutaneous administration of 400 mg/kg to rats on a gestational day 11 (Environmental Risk Assessment for Chemical Substances vol. 5 (Ministry of the Environment, 2006)). |

In an acute toxicity test by oral administration to rats (OECD TG423, GLP), at 300 mg/kg, transient loose stools were seen, but the animals showed smooth weight gain, and necropsy at the end of a 14-day observation period revealed no macroscopic changes in the visceral organs. At 2,000 mg/kg, decreased locomotor activity, deep and slow respiration, diarrhea, and soiled perineal region were observed, 3/3 animals died within 24 hours after a dose, no macroscopic changes were found at necropsy, and the approximate lethal dose was reported to be 500 mg/kg (JECDB (Access on May 2012)).
