It was classified in Category 3 based on an LD50 value of 500-1,000 mg/kg for guinea pigs (Initial Risk Assessment Report 208 (NITE, CERI, NEDO, 2005)). |

Warning |
H315 |
P302+P352
P332+P313 P362+P364 P264 P280 P321 |
It was classified in Category 2 because it is reported that severe irritation was observed in a 24-hour occlusive application test with guinea pigs (Hazard Assessment Report Ver. 1.0, No. 208 (CERI, NITE, 2005)), and it is described that this substance and its isomer mixture irritate the skin in humans and may cause erythema and pain on contact with the skin (Environmental Risk Assessment for Chemical Substances vol. 5 (Ministry of the Environment, 2006)). |
3 | Serious eye damage/eye irritation | Classification not possible |
- |
- | - | Data are lacking. Besides, it is described that this substance and its isomer mixture may irritate the eye (Environmental Risk Assessment for Chemical Substances vol. 5 (Ministry of the Environment, 2006)), but no data are quoted, and the details are unknown. |
4 | Respiratory sensitization | Classification not possible |
- |
- | - | No data available. |
4 | Skin sensitization | Classification not possible |
- |
- | - | No data available. |
5 | Germ cell mutagenicity | Classification not possible |
- |
- | - | Data are lacking. Besides, it was reported to be negative in a DNA synthesis inhibition test with the testis after oral administration to mice (in vivo germ cell genotoxicity test) (Environmental Risk Assessment for Chemical Substances vol. 7 (Ministry of the Environment, 2009)). And as for in vitro tests, Ames tests gave both positive (JECDB (Access on July 2012), NTP DB (Access on June 2012)) and negative results (Initial Risk Assessment Report 208 (NITE, CERI, NEDO, 2005)), and it was reported to be positive in a chromosomal aberration test with CHL/IU cells (JECDB (Access on July 2012)). |
6 | Carcinogenicity | Classification not possible |
- |
- | - | No data available. Besides, ACGIH classified it in A3 for carcinogenicity as mixed isomers. |
7 | Reproductive toxicity | Classification not possible |
- |
- | - | No data available. |
8 | Specific target organ toxicity - Single exposure | Classification not possible |
- |
- | - | It is described that exposure at high levels of this substance could cause lowering of consciousness and formation of methemoglobin (Environmental Risk Assessment for Chemical Substances vol. 7 (Ministry of the Environment, 2009)), but no specific data are quoted, and the details are unknown. Therefore, the classification is not possible due to lack of data. Besides, as data on 3,5-dimethylaniline, the isomer, it is reported that in an oral administration test with rats, blood methemoglobin level exceeded 5% after administration of 145 mg/kg or above (the control group, 4.96%) and reached the maximum of 32.7% at 3 hours after administration of 580 mg/kg (Environmental Risk Assessment for Chemical Substances vol. 7 (Ministry of the Environment, 2009)), and effects on the blood were observed. |
9 | Specific target organ toxicity - Repeated exposure | Category 2 (blood, kidney, liver) |
Warning |
H373 |
P260
P314 P501 |
In a 28-day repeated dose toxicity test by oral administration to rats (Guidelines specified by the Chemical Substances Control Law, GLP), decreases in hemoglobin level, erythrocyte count, and hematocrit value and increased methemoglobin were observed at or above 60 mg/kg (converted guidance value: 18.7 mg/kg/day) corresponding to the guidance value range for Category 2, histopathological examination revealed increased deposits of hemosiderin in the spleen and increased hematopoiesis in the bone marrow, and these were considered to be increased hematopoiesis to compensate for anemia with methemoglobinemia (JECDB (Access on July 2012)). Therefore, it was classified in Category 2 (blood). And at 300 mg/kg (converted guidance value: 93.3 mg/kg/day) corresponding to the guidance value range for Category 2, an increase in urinary protein, increases in erythrocytes and leukocytes and an increase in small round epithelial cells in urinary sediment, and an increase in urea nitrogen were found, and histopathological examination showed basophilic changes in the renal tubules and mineralization of the inner medulla, increased incidence of eosinophilic bodies in the renal tubular epithelium in addition to papillary necrosis, dilatation of the renal tubules, and cellular infiltration in the cortical interstitium (JECDB (Access on July 2012)). Therefore, it was classified in Category 2 (kidney). Furthermore, it was classified in Category 2 (liver) because increases in activities of gamma-GTP and GPT, an increase in total cholesterol, hypertrophy of centrilobular hepatocytes, and bile duct proliferation at the histopathological examination were seen at 300 mg/kg, and these were considered to be slight hepatic disorders (JECDB (Access on July 2012)). |
10 | Aspiration hazard | Classification not possible |
