P501
Among LD50 values for rats described in an assessment document (SIDS (2003)), two with higher reliability by SIDS (1,640 mg/kg bw and 3,740 mg/kg bw) were adopted for classification. Among them, one corresponds to Category 4, and the other corresponds to "Not classified." From a safety point of view, the category with higher hazard was adopted, and it was classified in Category 4.
1 Acute toxicity (Dermal) Not classified
-
-
- - It was classified as "Not classified" in the Classification JIS (corresponding to Category 5 in UN GHS classification) based on the one with higher reliability (4,920 mg/kg bw) among LD50 values for rabbits described in an assessment document (SIDS (2003)).
1 Acute toxicity (Inhalation: Gases) Not applicable

-
- - Liquid (GHS definition)
1 Acute toxicity (Inhalation: Vapours) Category 4

It was classified in Category 4 based on an LC50 value by 1-hour exposure for rats of 40.8-43.0 mg/L (converted 4-hour equivalent value: 4,307-4,538 ppm) (SIDS (2003)). Besides, because the toxicity value (40.8-43.0 mg/L) was lower than 90% of the saturated vapour pressure concentration (77 mg/L), a reference value of gasses was applied. Besides, there is a report on an LC50 value by 4-hour exposure for rats of > 6.1 mg/L (> 1,310 ppm) (SIDS (2003)), but the category cannot be determined with the data.
1 Acute toxicity (Inhalation: Dusts and mists) Classification not possible
-
-
- - No data available.
2 Skin corrosion/irritation Category 2

In a test by a 4-hour occlusive application of 0.5 mL of the undiluted test substance to the skin of six rabbits, the average scores at 24, 48, 72 hours after removal of the patch were 2.0, 2.5, 2.5 for erythema and 3.3, 2.7, 2.3 for edema (SID (2003)). Because the average scores for edema were >= 2.3 and <= 4.0 on three consecutive days, it was classified in Category 2.
3 Serious eye damage/eye irritation Not classified
-
-
- - In a test in which 0.1 mL of undiluted this substance was applied to the conjunctival sac of rabbits (OECD TG405), mild to moderate conjunctivitis occurred 1 hour after the application, the average scores after 24, 48, 72 hours were 1.0, 0.8, 0.2 for conjunctival redness, 0.3, 0, 0 for chemosis, and all 0 for cornea and iris (SIDS (2003)). From the above, because all the average scores for three days were less than 1 for the conjunctiva, iris, and cornea, it was classified as "Not classified."
4 Respiratory sensitization Classification not possible
-
-
- - No data available.
4 Skin sensitization Classification not possible
-
-
- - Data are lacking. Besides, it is described that it was not sensitizing in a sensitization test with guinea pigs (Epicutaneous test) (IUCLID (2000)), but the test details are unknown.
5 Germ cell mutagenicity Not classified
-
-
- - It was classified as "Not classified" based on negative results in micronucleus tests with bone marrow cells after oral or inhalation exposure in mice (in vivo somatic cell mutagenicity test) (OECD TG474, GLP) (SIDS (2003)). Besides, as for in vitro tests, it is reported that it was positive in an Ames test, and an HGPRT test with cultured cells of Chinese hamsters was negative in CHO cells and positive in V79 cells (SIDS (2003)).
6 Carcinogenicity Classification not possible
-
-
- - No data available.
7 Reproductive toxicity Classification not possible
-
-
- - No data available.
8 Specific target organ toxicity - Single exposure Category 2 (nervous system), Category 3 (narcotic effects, respiratory tract irritation)

Sluggishness, prostration, moribund appearance, lacrimation, unsteady gait, and tremors were observed in the groups of 1,000 mg/kg or above in an acute oral toxicity test with rats (LD50: 1,640 mg/kg) (SIDS (2003)), and apathy, ataxia, and closed eyes in the 1,000 mg/kg dose group, and apathy, ataxia, prostration, reduced or no reflexes, and labored breathing in the groups of 2,150 mg/kg or above were found in another acute oral toxicity test with rats (LD50 value: 3740 mg/kg) (SIDS (2003)). From the above, after oral administration to rats, neurobehavioral symptoms were seen at 1,000 mg/kg corresponding to the guidance value range for Category 2, and narcosis-like actions such as apathy and reduced reflexes were observed in the groups of 2,150 mg/kg or above. Therefore, it was classified in Category 2 (nervous system), Category 3 (narcotic effects). And it was classified in Category 3 (respiratory tract irritation) because it is described that there were no deaths in an inhalation test by exposure to vapour in rats (dose: 6.1 mg/L/4 hours), but irritation to the respiratory tract was found (SIDS (2003)).
9 Specific target organ toxicity - Repeated exposure Category 1 (respiratory tract)

P501
In a 2-week repeated inhalation exposure test with rats (vapour: 6 hours/day, total 9-time exposure) (GLP-compliant), findings were edema in the nasal cavity and squamous metaplasia in the respiratory mucosa in the low dose group (480 mg/m3, converted guidance value: 0.075 mg/L), mild to moderate rhinitis, cell degeneration or atrophy of the olfactory epithelium, squamous metaplasia in the epithelium of the nasal cavity and to a lesser degree in the larynx and trachea in the middle dose group (4,660 mg/m3, converted guidance value: 0.725 mg/L), and signs of respiratory distress, respiratory tract injury by histological examination, and deaths of all animals (30/30) in the high dose group (20,640 mg/m3, converted guidance value: 3.21 mg/L) (SIDS (2003)). From the above, because adverse effects on the respiratory tract were observed at or above the low dose (480 mg/m3, converted guidance value: 0.075 mg/L) corresponding to the guidance values for Category 1, it was classified in Category 1 (respiratory tract).
10 Aspiration hazard Classification not possible

-
- - No data available.

Rationale for the classification
11 Hazardous to the aquatic environment (Acute) Not classified
-
-
- - It was classified as "Not classified" because toxicity values are > 100 mg/L for all of the algae, crustacea, and fish (SIDS, 2005).
11 Hazardous to the aquatic environment (Long-term) Not classified
-
-
- - Reliable chronic toxicity data were not obtained. It was classified as "Not classified" because it is not water-insoluble (water solubility: 18800 mg/L, PHYSPROP Database (2009)), and it corresponds to "Not classified" in acute toxicity for algae, crustacea, and fish (SIDS, 2005).
12 Hazardous to the ozone layer Classification not possible
-
-
- - This substance is not listed in the Annexes to the Montreal Protocol.
