1 | Acute toxicity (Oral) | Category 4 |
Warning |
H302 |
P301+P312
P264 P270 P330 P501 |
There is a report on two LD50 values of 1,800 mg/kg and 2,280 mg/kg for rats (SIDS (Access on June 2012)), corresponding to Category 4 and "Not classified" in the Classification JIS (corresponding to Category 5 in UN GHS classification), respectively. Therefore, it was classified in Category 4 by adopting a more hazardous category. |
1 | Acute toxicity (Dermal) | Not classified |
- |
- | - | It was classified as "Not classified" (corresponding to Category 5 in UN GHS classification or "Not classified") based on an LD50 value of > 2,000 mg/kg for rabbits (SIDS (Access on June 2012)). |
1 | Acute toxicity (Inhalation: Gases) | Not applicable |
- |
- | - | Liquid (GHS definition) |
1 | Acute toxicity (Inhalation: Vapours) | Classification not possible |
- |
- | - | There is a report on an LC50 value by 4-hour exposure of >= 21.2 mg/L = 6,018 ppm for rats (OECD TG 403) (SIDS (Access on June 2012)), but because the category cannot be determined with the data alone, the classification is not possible. Besides, because the LC50 value (21.2 mg/L) was lower than 90% of the saturated vapour pressure concentration (109 mg/L), a reference value of gas was applied. |
1 | Acute toxicity (Inhalation: Dusts and mists) | Classification not possible |
- |
- | - | No data available. |
2 | Skin corrosion/irritation | Not classified |
- |
- | - | It was classified as "Not classified" because it was not irritating in a skin irritation test with rabbits (SIDS (Access on June 2012)). |
3 | Serious eye damage/eye irritation | Not classified |
- |
- | - | It was classified as "Not classified" because it was not irritating in an eye irritation test with rabbits (SIDS (Access on June 2012)). |
4 | Respiratory sensitization | Classification not possible |
- |
- | - | No data available. |
4 | Skin sensitization | Classification not possible |
- |
- | - | It is reported that it was not sensitizing in a maximization test with guinea pigs (Directive 84/449/EEC, B.6, GLP) (IUCLID (2000)), but it is List 2 data, and the test details such as positive rate are unknown. Therefore, it was classified as "Classification not possible." |
5 | Germ cell mutagenicity | Not classified |
- |
- | - | It was classified as "Not classified" based on a negative result in a micronucleus test by oral administration to mice (in vivo mutagenicity test) (OECD TG 474, GLP) (SIDS (Access on June 2012)). Besides, as for in vitro tests, it was reported to be negative in an Ames test (SIDS (Access on June 2012)). |
6 | Carcinogenicity | Classification not possible |
- |
- | - | No data available. |
7 | Reproductive toxicity | Category 2 |
Warning |
H361 |
P308+P313
P201 P202 P280 P405 P501 |
In a one-generation reproductive test with rats orally administered before mating and during the organogenesis period (OECD TG 421 (Draft), GLP), body weights of parent animals tended to decrease or slightly decreased in the high dose group (200 mg/kg/day), but there were no differences in parameters for mating, fertility, and reproduction between treated groups and the control group. And as for the development of offspring, no abnormal findings were observed at the morphological examination of external, visceral, and skeletal system at the dose. However, because the pup viability index was reduced at the high dose, shown by an increased number of pups dying during lactation and a decreased number of pups surviving on day 5 of lactation (SIDS (Access on June 2012)), it was classified in Category 2. |
8 | Specific target organ toxicity - Single exposure | Category 3 (Narcotic effects) |
Warning |
H336 |
P304+P340
P403+P233 P261 P271 P312 P405 P501 |
In a micronucleus test by single oral administration of 500, 1,000, and 1,500 mg/kg to mice (OECD TG 474, GLP), signs of irregular respiration, piloerection, and apathy were observed at or above 1,000 mg/kg (SIDS (Access on June 2012)), and in a 28-day repeated oral administration test with rats (OECD TG 407, GLP), there is a report on sedation, ataxia, and uncoordinated gait as toxic signs at 600 mg/kg during the first day of treatment (SIDS (Access on June 2012)). Therefore, it was classified in Category 3 (narcotic effects). |
9 | Specific target organ toxicity - Repeated exposure | Classification not possible |
- |
- | - | Two 28-day repeated oral administration tests with rats (OECD TG 407, GLP) are reported. In one test, apparent effects on the liver and kidney were observed at or above 200 mg/kg/day (converted guidance value: 62.2 mg/kg/day) and were hypertrophy of hepatocytes in the liver and accumulation of hyaline droplets in the kidney. Liver findings were not regarded as adverse effects because they were not accompanied by changes in related blood biochemical values, and it is described that kidney findings were alpha-2micro-Globulin accumulation specific to male rats and not applicable to humans (SIDS (Access on June 2012)). In the other test, symptoms such as apathy and salivation and changes in blood biochemical values such as decreased cholesterol were observed in some animals at 750 mg/kg/day (converted guidance value: 233 mg/kg/day), but there were no treatment-related changes in a histopathological examination, and no effects of administration were found at or below 150 mg/kg/day (IUCLID (2000)). From the above, because no adverse effects of administration were seen at doses within the guidance value range, it corresponds to "Not classified" in the oral route. However, due to no data on other routes, it was classified as "Classification not possible" for specific target organ toxicity (repeated exposure). |

Rationale for the classification | |
---|---|---|---|---|---|---|
11 | Hazardous to the aquatic environment (Acute) | Not classified |
- |
- | - | It was classified as "Not classified" because toxicity values are > 100 mg/L for all od the algae, crustacea, and fish (SIDS, 2005). |
11 | Hazardous to the aquatic environment (Long-term) | Not classified |
- |
- | - | Reliable chronic toxicity data were not obtained. It was classified as "Not classified" because it is not water-insoluble (water solubility: 1000000 mg/L, PHYSPROP Database (2009)), and it corresponds to "Not classified" in acute toxicity for algae, crustacea, and fish (SIDS, 2005). |
12 | Hazardous to the ozone layer | Classification not possible |

- | - | This substance is not listed in the Annexes to the Montreal Protocol. |
