There are many reports of micronucleus tests with bone marrow or peripheral blood after oral or inhalation administration to mice (in vivo somatic cell mutagenicity tests), most of which gave positive results (JECFA 1125 (2006)). Therefore, it was classified in Category 2. Besides, as for other in vivo tests, a dominant lethal test with mice and a specific locus test with mice (in vivo heritable germ cell mutagenicity tests) were reported to be negative (JECFA 1125 (2006)), and a chromosomal aberration test, a sister chromatid exchange test, and an unscheduled DNA synthesis test with somatic cells were reported to be positive (JECFA 1125 (2006)). As for in vitro tests, it is reported that it was negative or positive in Ames tests, positive in a chromosomal aberration test with human peripheral lymphocytes, and negative in a chromosomal aberration test with CHO cells (Initial Risk Assessment Report, Ministry of Health, Labour and Welfare (Access on August 2011), JECFA 1125 (2006), NTP DB Study ID 588195 (1987)). |

In a test by inhalation exposure of mice on a gestational day 9, significant increases in external anomalies (cleft palate, polydactyly) and abnormal spines were reported in fetuses after 24-hour or 36-hour exposure (Initial Risk Assessment Report, Ministry of Health, Labour and Welfare (Access on August 2011)), although there are no descriptions of general toxicity in maternal animals. Furthermore, after oral administration of 1,000 mg/kg to pregnant rats, decreased food consumption and reduced weight gain were observed as general toxicity in maternal animals, on the other hand, as reproductive effects, in addition to decreased body weight and increased resorptions in fetuses, malformations of absent tails and exencephaly brain were seen after administration on gestational days 6-12, and increases in skeletal abnormalities in the vertebrae and sternebrae were found after administration on gestational days 0-5, 6-12, or 13-19 (JECFA 1125 (2006)). In 13-week repeated oral administration tests (drinking water) with rats and mice, in rats, decreases in leukocytes and lymphocytes at or above 11-23 mg/kg/day and lymphoid depletion in the spleen and lymph nodes at or above 78-114 mg/kg/day, corresponding to the guidance values for Category 2, were observed, and although the doses were above the guidance value range, a significant decrease in bone marrow cell counts in rats and lymphoid depletion in the spleen, lymph nodes, and thymus and a decrease in bone marrow cell counts in mice were found (Initial Risk Assessment Report, Ministry of Health, Labour and Welfare (Access on August 2011)). Based on the above report, it was classified in Category 2 (hematopoietic system).
