In a two-generation reproductive toxicity test by diet administration prior to mating in male and female rats, hepatocellular vacuolization and hypertrophy at 100 ppm (7.3 mg/kg/day) or higher in parent animals as general toxicity, decreased fertility and decreased survival in gestation with a decrease in litter size through two generations, changes in sex cycle accompanied by prolonged estrus, and decreased neonatal survival at the highest concentration of 1000 ppm (74 mg/kg/day) were also observed. In a 3-month diet administration test in mice, dose-dependent elevated alkaline phosphatase and centrilobular hypertrophy of hepatocytes were observed at doses of 450 ppm (67.5 mg/kg/day) or higher. In a 2-year diet administration test in rats, increased cholesterol and triglycerides, fatty changes in livers, and eosinophilic changes of hepatocytes were observed at 1000 ppm (41.2 mg/kg/day). (IRIS (2002))

On the other hand, in a 90-day or 1-year diet administration test in dogs, degeneration in zona fasciculata and zona reticularis of adrenal cortex characterized by eosinophilic degeneration, vacuolation and cortical atrophy in animals at doses of 30 mg/kg/day or higher were reported as histopathological signs.
