Based on positive results in an inhalation rat and a mouse erythrocyte micronucleus test (in vivo somatic cell mutagenicity tests) (SIDS (2004)), the substance was classified into Category 2. As relevant information, from in vitro mutagenicity tests, there are reports of a negative Ames test (SIDS (2004)) and a negative rat liver cell chromosome aberration test (IUCLID (2002)).

7 Reproductive toxicity Classification not possible - - - - In a combined repeat dose and reproductive/developmental screening rat inhalation test (OECD 422 (GLP)), the estrus cycle was unaffected by exposure, mating performance and fertility indices were similar in all groups. Additionally, no evidence of reproductive or developmental toxicity was seen (SIDS (2004)). In a rat inhalation test (vapour) (GLP) there were no deaths and macroscopic and microscopic examination at necropsy after exposure was terminated revealed no compound related effects at 61000 ppm/4h. The animals became narcotized but revived within 30 minutes of cessation of exposure (SIDS (2004)). Additionally, there is a report of lethargic as a sign after oral administration to rats (SIDS (2004)). Therefore, the substance was classified into Category 3 (narcotic effects).
9 Specific target organ toxicity - Repeated exposure Classification not possible - - - - In a combined repeat dose and reproductive/developmental test (OECD 422 (GLP)) where rats were exposed via inhalation (vapour) for 28 days, decreased incidence of extramedullary haemopoiesis of the spleen, increased globlet cell hyperplasia in the nasal passages of males and minimal centrilobular hepatocyte hypertrophy in the liver of females were noted at a dose level of 7000 ppm/6h (20 mg/L/6h) (90-day equivalence: 6.25 mg/L/6h) which exceeds the guidance value range of Category 2 (<=1.0 mg/L/6 hr). In the 2000 ppm (5.2 mg/L/6h) (90-day equivalence: 1.6 mg/L/6h) dosed males, a slight increase in severity of myocardial inflammatory heart lesions and cortical/medullary tubular basophilia in the kidneys were observed (SIDS (2004)). Classification was not possible due to lack of data for evidence of effects near the upper limit guidance value range of Category 2.
