Based on a LC50 value (1 hour) of 163 ppm (4-hour equivalent: 81.5 ppm) for mice (HSDB (2002)), the substance was classified into Category 1. Since the LC50 value was lower than 90% of saturated vapour pressure concentration (62368.4 ppmV), the classification criteria for gas (ppm) was adopted.
1 Acute toxicity (Inhalation: Dusts and mists) Classification not possible - - - - No data available.
2 Skin corrosion/irritation Not classified - - - - Based on descriptions of "not caused severe damage" in a rabbit skin application test and "slightly irritating" in a guinea pig test (PATTY (5th, 2001)), the substance was classified as "Not classified" in JIS Classification (corresponding to Category 3 in GHS classification). In addition, there is a result of "mild irritating" in a Draize test where 500 mg was applied to rabbits for 24-hour (RTECS (2002)).
3 Serious eye damage/eye irritation Category 2 Warning H319: Causes serious eye irritation P305+P351+P338: IF IN EYES: Rinse cautiously with water for several minutes. Remove contact lenses, if present and easy to do. Continue rinsing.
P337+P313: If eye irritation persists: Get medical advice/attention.
P264: Wash ... thoroughly after handling.
P280: Wear protective gloves/protective clothing/eye protection/face protection.
Based on reports of "irritating to eye" as human health effects (HSDB (2002)) and "not caused severe damage" in a rabbit eye application test (PATTY (5th, 2001)), the substance was classified into Category 2. As relevant information, there is a result of "moderate irritating" in a rabbit Draize test (RTECS (2002)).
4 Respiratory sensitization Classification not possible - - - - No data available.
4 Skin sensitization Classification not possible - - - - No data available.
5 Germ cell mutagenicity Classification not possible - - - - Classification not possible due to lack of data from in vivo mutagenicity tests. From in vitro mutagenicity tests, there are reports of a negative Ames test, a negative CHO cells chromosomal aberration test and a negative sister chromatid exchange analysis (NTP DB (access on Sep. 2009)).
6 Carcinogenicity Not classified - - - - From Carcinogenicity Studies (Ministry of Health, Labour and Welfare) (2006), there is a result that "In a 104-week inhalation test with rats and mice, exposure of propanenitrile did not increase the incidence of any tumor in male or female F344/DuCrj (Fischer) rats and then there was no evidence of carcinogenic activity. For Crj:BDF1 mice, there was no evidence of carcinogenic activity since no increase in incidence of any tumor was observed in either sex". Therefore the substance was classified as "Not classified".
7 Reproductive toxicity Category 1B Danger H360: May damage fertility or the unborn child P308+P313: IF exposed or concerned: Get medical advice/attention.
P201: Obtain special instructions before use.
P202: Do not handle until all safety precautions have been read and understood.
P281: Use personal protective equipment as required.
P405: Store locked up.
P501: Dispose of contents/container to ...
Based on a report that embryonic death and fetal toxicity were observed in an inhalation developmental toxicity test with rats at a dose at which general toxicity was not manifested in parental animals (PATTY (5th, 2001)), the substance was classified into Category 1B.
8 Specific target organ toxicity - Single exposure Category 1 (systemic toxicity, central nervous system) Danger H370: Causes damage to organs (systemic toxicity, central nervous system) P307+P311: IF exposed: Call a POISON CENTER or doctor/physician.

Since there are human case reports that a worker exposed to the substance via inhalation and dermal contact showed loss of consciousness, respiratory alkalosis and metabolic acidosis, and that 2 workers inhaled the substance with 1 worker exhibiting symptoms of headache, nausea and dizziness 2-hour after exposure and the second worker collapsing 7-hour after exposure (PATTY (5th, 2001)), the substance was classified into Category 1 (systemic toxicity, central nervous system).
9 Specific target organ toxicity - Repeated exposure Classification not possible - - - - Although there is report that changes in urine composition and erythrocyte count, weight loss or decreased weight gain were observed at 209 ppm (0.47 mg/L) in a mouse 14-week inhalation test (RTECS (2002), original literature: Toxicologist. (Conference Paper in Society of Toxicology) Vol. 5, p.113, 1985), classification was not possible due to lack of the detailed data. As relevant information, the respiratory system, cardiovascular system, central nervous system, liver, kidneys are listed as target organ in NIOSH Pocket Guide to Chemical Hazards (2005).
10 Aspiration hazard Classification not possible - - - - No data available.

ENVIRONMENTAL HAZARDS

11 Hazardous to the aquatic environment (Acute) Not classified - - - - Classified into Not classified from its 96h-LC50 = 1520 mg/L for fish (Fathead minnow) (HSDB, 2002; AQUIRE, 2010).
11 Hazardous to the aquatic environment (Long-term) Not classified - - - - Classified into Not classified since its acute toxicity is Not classified and it is not non-water soluble from water solubility = 100,000 mg/L (PHYSPROP Database, 2010).
