In 4-hour exposure tests, the rat LC50 values were 0.27mg/L (OECD TG 403) and > 0.52mg/L (OECD TG 403) (IUCLID, 2000). Of two LC50 values, 0.27mg/L, indicating higher toxicity, was used as a basis by which the substance was classified into Category 2. The EU has classified the substance into R23.
2 Skin corrosion/irritation Category 2 Warning H315: Causes skin irritation P302+P352: IF ON SKIN: Wash with plenty of soap and water.
P332+P313: If skin irritation occurs: Get medical advice/attention.
P264: Wash ... thoroughly after handling.
P280: Wear protective gloves/protective clothing/eye protection/face protection.
P321: Specific treatment (see ... on this label).
P362: Take off contaminated clothing and wash before reuse.
The substance was found not to be irritating in human tests (IUCLID, 2000). On the other hand, rabbit tests yielded contradictory results: the substance caused severe irritation in one test and was not irritating in another (IUCLID, 2000). Based on the EU's classification of the substance into R36, 37, and 38 (EU-Annex 1, accessed in July 2008), it was classified into Category 2 taking the higher risk rating by the EU into account.
3 Serious eye damage/eye irritation Category 2A Warning H319: Causes serious eye irritation P305+P351+P338: IF IN EYES: Rinse cautiously with water for several minutes. Remove contact lenses, if present and easy to do. Continue rinsing.
P337+P313: If eye irritation persists: Get medical advice/attention.
P264: Wash ... thoroughly after handling.
P280: Wear protective gloves/protective clothing/eye protection/face protection.

4 Respiratory sensitization Category 1 Danger H334: May cause allergy or asthma symptoms or breathing difficulties if inhaled P304+P341: IF INHALED: If breathing is difficult, remove victim to fresh air and keep at rest in a position comfortable for breathing.
P342+P311: If experiencing respiratory symptoms: Call a POISON CENTER or doctor/physician.
P261: Avoid breathing dust/fume/gas/mist/vapours/spray.
P285: In case of inadequate ventilation wear respiratory protection.
P501: Dispose of contents/container to ...
Occupational asthmatic patients showed positive reactions in bronchus provocation tests (IUCLID, 2000), and the substance has been classified into R42 in EU classification (EU-Annex 1 (accessed in July 2008)). Thus, the substance was classified into Category 1.
4 Skin sensitization Classification not possible - - - - No data available.
5 Germ cell mutagenicity Classification not possible - - - - In vitro mutagenicity tests (Ames tests) yielded negative results (IUCLID, 2000). However, since there was no in vivo data available, classification is not possible.
6 Carcinogenicity Not classified - - - - Since IARC classified the substance into Group 3 in carcinogenicity assessment (IARC 71 (1999)), it was classified into "Not classified".
7 Reproductive toxicity Classification not possible - - - - No data available.
8 Specific target organ toxicity - Single exposure Category 3 (Respiratory tract irritation) Warning H335: May cause respiratory irritation (respiratory tract irritation) P304+P340: IF INHALED: Remove victim to fresh air and keep at rest in a position comfortable for breathing.
P403+P233: Store in a well-ventilated place. Keep container tightly closed.
P261: Avoid breathing dust/fume/gas/mist/vapours/spray.
P271: Use only outdoors or in a well-ventilated area.

P405: Store locked up.
P501: Dispose of contents/container to ...
The link between exposure to this substance and bronchitis has been documented based on observations that workers who handled this substance at the polyurethane rubber producing plants suffered from conjunctival irritation in addition to acute disorder in the respiratory tracts. In addition, as a result of epidemiological studies on rubber production workers, the substance has been confirmed to be an irritant since exposure to it was associated with increased frequencies of eye irritation, coughing, and exertional dyspnea. Moreover, increased closing volume seen particularly in elderly workers in respiratory function tests is considered to be an indicator of respiratory tract irritation. Based on these human cases, the substance was classified into Category 3 (respiratory tract irritation). By the EU, the substance has been classified into R36, 37, and 38 (EU-Annexl (accessed July 2008)).
9 Specific target organ toxicity - Repeated exposure Category 2 (respiratory system) Warning H373: May cause damage to organs through prolonged or repeated exposure (respiratory system) P260: Do not breathe dust/fume/gas/mist/vapours/spray.
P314: Get medical advice/attention if you feel unwell.
P501: Dispose of contents/container to ...
There is a case report in which a worker who handled polyurethane containing this substance repeatedly suffered from high fever and seizures accompanied with severe respiratory symptoms three times within a month, and died during seizures (IUCLID (2000)). However, this worker did not show any symptoms while handling the same material prior to the incident (IUCLID (2000)). In addition, because some workers (unidentified number) who handled materials containing this substance complained of bronchitis, an observational study was conducted for 1 year. This study indicated that there may be a positive relationship between isocyanate and thoracic disorder (IUCLID (2000)). Based on these List 2 assessment documents on human cases, the substance was classified into Category 2 (respiratory system).
10 Aspiration hazard Classification not possible - - - - No data available.
