1 Acute toxicity (Inhalation: Vapours) Classification not possible - - - - Saturated vapor pressure level at 20degCis 223 ppm. It is written in GESTIS (Access on Feb.2008) of Priority 3 that there was no mortality and no clinical symptom in case that rats were exposed in the saturated steam at 20degC for 7 hours (4-hour equivalent 0.71 mg/L). Since the vapor standard value of Category 2 is 2.0 mg/L, classification is not possible.

Based on the statement of the "eye irritation" of ICSC (2001) and HSDB (2002), and the statement of HSDB (2002) of "the onset of conjunctivitis is indicated and the effect is mild in animals studies," it is classified into Category 2B. In addition, GESTIS (Access on Feb.2008) of Priority 3 indicates as "Since the test result to eyes of rabbits varies, it is necessary to take into consideration the reversible damage at the time of contacting human's eyes directly".

There is a description as follows in HSDB (2002), "With dermal administration to female rats of 7-14 day prgnancy 87% of fetuses died and deformation (hydronephrosis, hydrocephalus) was caused to all live fetuses and similar results were obtained via the oral route. In the case of dermal route applied to pregnant rabbits during the formation period of fetal organs a low level of teratogenicity was observed." When content of the primary document of HSDB was closely reviewed, the effect found in pregnant rats within the range of dose concentrations was that causing the body weight reduction (7-9%). Effects on rabbits are not known because their body weight was not measured. Since fetal death and teratogenicity were found with rats after all at the concentration level where almost no maternal toxicity exsisted, it was classified into Category 1B. EU Annex 1, however, corresponds to Cat.2, R61, corresponding to GHS Category 1B.

Although ICSC (2001) of Priority 2 has a description of "cough", it dose not mention any irritation other than eye irritation in the paragraph of short term exposure. Moreover, to GESTIS of Priority 3 (Access on Feb.2008) has a description that "there is oral administration data solely as a cancer drug, and nausea, vomiting, and an impaired liver function were seen by the concentration beyond 15 mg/kg which is not so high" on humans. Although human data is based on the information source of Priority 2,3, since it can be predicted that there is abundance of clinical cases as clinical drugs, it is defined as Category 1 (liver, systemic symptoms, respiratory system).

Short term exposure has effects on liver (ICSC (2001)). As for human, HSDB (2002) shows that "the patients who were administered the substance as an oral cancer drug experienced nausea, vomiting and poor appetite, and those who were administered a total of 80 mg/kg or more were found to have damage in the liver in the liver function test" and there is the description in the Priority 3 of GESTIS that "although most human exposure information is based on the oral administration as cancer drugs, a short-term exposure to much smaller concentrations causes remarkable disorder in respiratory function." Since the check of the primary literature of GESTIS is difficult, the specific period is unknown. Although human data is based on the information source of Priority 2,3, since it can be predicted that there is abundance of clinical cases as clinical drugs, it is defined as Category 1 (liver, systemic symptoms, respiratory system).
