Although there is no skin irritation test data of this substance, it has been described that this substance has a "skin irritation" in ICSC (2001) or that "Diaminotoluene irritates skin" in EHC 74 (1987). It may be classified as Category 2 or Category 3 since it is likely to have an irritation despite its unknown degree. It is classified into Category 2 from a viewpoint of safety. In addition, 2,5-TDA and 2,4-TDA can be classified as Category 3.

Although there is no data of the eye irritation test of this substance, it has been described that the substance is "eye-irritative" in ICSC (2001), or that "Diaminotoluene irritates eyes" in EHC 74 (1987). Although the level is unknown, since it is considered to own irritation, it is classified into Category 2. In addition, 2,5-TDA is Category 2A-2B and 2,4-TDA are Category 2A.

Moreover, although 2,4-TDA is defined as "Not classified" as a result of the previous classification, a positive result has been obtained in the above mutation assays employing transgenic mice, and it is classified into Category 2.
6 Carcinogenicity Not classified - - - - There is no existing classification. On the other hand, EHC 74 (1987) cited the 103-week oral administration test employing rats and mice by NCI (1980). EHC is saying that "NCI (1980) concluded that 2,6-TDA does not show carcinogenicityity to F344 rat of male and female, and B6C3F1 mouse of male and female" based on various statistical evaluations, and the reliability of the data of NCI can be evaluated. Therefore, the conclusion of NCI was adopted and it was carries out "Not classified". In addition, in a three-ministry GHS classification, 2,5-TDA is "Not classified", 2,4-TDA and TDA are in Category 2.

Since embryonic death has been identified at the dose level where decrease in the body weight of mother animals is identified in the oral administration study using rabbits in the description of EHC 74 (1987), it is classified in Category 2. In addition, since increase of imperfect spinal segments development in fetuses at the dose level where the body weight reduction of mother animals is not identified is identified in the oral administration study using rats published in EHC 74 (1987), which corresponds to a skeletal variant, GHS is considered to correspond to Category 2. Moreover, strenebrae deficiencies and imperfect closure of skulls have been observed at the dose level where the body weight reduction in mother animals is observed. 2.5-TDA, 2.4-TDA and TDA are all Category 2.

About humans, ICSC (J) (2001) which is the information source of Priority2, have descriptions of "cough, dizziness, headache, shortness of breath, confusion, spasm, nausea, and deliquium animi" as acute symptoms by inhalation exposures, and that "airway is stimulated. Blood may be affected and a methemoglobin may be generated" as effects of short durations exposures. Therefore, it was considered to have Respiratory tract irritation and Narcotic effects, and its target organ was the blood systems. As mentioned above, the category was taken as Category 2 (blood system) and Category 3 (Respiratory tract irritation, Narcotic effects).
