Based on the evidence of "slight congestion in the skin, moderate skin necrosis and incrustation" (EHC 163, 1994) from the rabbit skin irritation test, although the substance should be placed in Category 1A from the viewpoint of safety.

Based on the evidence of "Severe eye irritation, with mydriasis and keratitis in all rabbits. Translucent zones in the cornea were observed in four animals and a purulent hemorrhagic discharge was also reported (number of rabbits unknown). The effects had disappeared 2-3 weeks after application, except for one rabbit that still showed corneal opacity after 3 weeks" (EHC 163, 1994) from the rabbit eye irritation test.

Based on the absence of data on heritable mutagenicity tests, germ cell mutagenicity tests in vivo and germ cell genotoxicity tests in vivo and positive data on somatic cell mutagenicity tests in vivo (micronucleus and chromosome aberration tests), described in CERI-NITE Hazard Assessment No.16 (2004), IARC 73 (1999), CaPSAR (2001), EHC 163 (1994), DFGOT Vol. 14 (2000) and NTP DB (Access on December 2005).

Due to the fact that the substance is classified as Category R by NTP (2005), Group 2B by IARC (1999), Category 3 by EU Risk Phrase, Category A3 by ACGIH (2001), Category B2 by EPA (1986) and Category 2B by the Japan Society for Occupational Health.
7 Reproductive toxicity Category 2 Warning H361: Suspected of damaging fertility or the unborn child P308+P313: IF exposed or concerned: Get medical advice/attention.
P201: Obtain special instructions before use.
P202: Do not handle until all safety precautions have been read and understood.
P281: Use personal protective equipment as required.
P405: Store locked up.
P501: Dispose of contents/container to ...
Based on the evidence of a decline in fertility, a decreas in crown-rump length, delayed calcification of the skull and lumbar ribs, an increase in cleft palate, malformation of the interperietal bone, increased incidence of anuary, brachyury and anal atresia within a litter, subcutaneous edema and increased rates of absorbed embryos at dosing levels toxic to parent animals in mouse three-generation tests and rat and mouse teratogenicity tests, described in CERI-NITE Hazard Assessment No.16 (2004) and IRIS (2001). Although an increase in conception rates in the F1 generation (CERI-NITE Hazard Assessment No.16, 2004), an increase in epididymidis weight and degeneration of duct epithelium of the cauda epididymidis (IRIS, 2001) were observed at dosing levels not toxic to parent animals in the mouse teratogenicity tests, and incomplete ossification of the skull (CERI-NITE Hazard Assessment No.16, 2004) was observed at dosing levels not toxic to parent animals in rabbit teratogenicity tests, the dosing level affecting the epididymis was not specified, and the rest of the changes seemed insignificant. Thus, placement of the substance in Category 2 seems appropriate.

P501: Dispose of contents/container to ...
Although acute toxicity was Category 2 and the bio-accumulation potential was low (BCF=13(Existing Chemical Safety Inspections Data)), since there was no rapidly degrading (the decomposition by BOD: 0%(Existing Chemical Safety Inspections Data)), it was classified into Category 2.
